Description

BT93/6 / Spore Coupon Biological Indicator for monitoring Plasma or Vaporized Hydrogen Peroxide processes. Geobacillus stearothermophilus (ATCC® 7953) 10^6 spores per coupon.

The envelope containing Bionova® BT93 Spore Coupon is specifically designed to control Plasma or Vaporized Hydrogen Peroxide sterilization processes. Spore coupons should be incubated in MC1020-2 Bionova® Growth Medium tube or other medium appropriate for Geobacillus stearothermophilus ATCC® 7953 growth.

If sterilization process was not effective, MC1020-2 Growth Medium will turn to yellow after incubation between 55-62 ºC, thus indicating the existence of living Geobacillus stearothermophilus spores in the spore coupon.

If sterilization process was effective, MC1020-2 Growth Indicating medium will remain its original color. Final readout should be done after a 24-hour incubation between 55-62 ºC of the growth medium containing the spore coupon.

Instructions for use

1. Pack the envelope in an appropriate package along with items for sterilization, according to recommended sterilization practices. Place it in those areas you consider a priori more inaccessible to the sterilizing agent. The load center is generally a problematic area.

2. Sterilize as usual.

3. After sterilization process has finished, remove Bionova® BT93 envelope from the package for processing and incubation.

4. Tear the envelope at one end very carefully in sterility conditions (eg. laminar flow cabinet) and transfer the spore coupon using a sterile clamp to MC1020-2 Growth Mediumtube, or any other appropriate growth medium.

WARNING: Avoid spore coupon contact with any kind of surface including the outer part of growth medium tube and Bionova® BT93 envelope.

IMPORTANT: Use latex gloves and a mask when transferring spore coupons from the envelope to the Growth Medium tube.

Use a spore coupon which has not been subjected to sterilization process as a positive control whenever you incubate a processed coupon. Both, processed spore coupon and the coupon used as positive control must belong to the same batch. Positive control guarantees that the incubation was carried out under appropriate conditions.

5. Incubate processed spore coupon along with the coupon used as positive control during a maximum of 24 hours between 55-62 ºC. It is advisable to make observations every 10 hours. Color change of MC1020-2 to yellow means a failure in sterilization process.

If after 24 hours no color change is visible on MC1020-2 Growth Medium, the result is negative (i.e. the sterilization process was effective. MC1020-2 growth medium used for incubating the spore coupon which have not been sterilized (positive control) should turn to yellow for the results to be valid.
